Abstract | Transcription initiation in bacteria can be divided in three main processes: RNA polymerase binding to the promoter in the DNA, followed by open complex formation and then promoter escape. The efficiency of transcription initiation therefore depends on the efficiency with which RNA polymerase binds the promoter, on the rate of open complex formation, and on the efficiency of promoter escape. The rates at which those three processes occur are not independent. For example, the rates of promoter escape are different for different RNA polymerase binding affinities for the promoter. In this talk I will present our approach to promoter escape using Markov chains. |
